It looks like you turn the radio on while pressing the M/CALL-PRIO button. When the computer is ready, then press the SET-LOCK button to send the data into the computer or other radio. Reading this, it looks like you may not need to do anything if you are sending data to the radio, as long as the programming cable is connected to the radio and computer. If you use Icom's OPC-478U USB-cable and CS-208 programming software you don't have to do anything.
Just plug the cable into the external speaker jack on the radio and into a USB connector on your computer, turn on the radio, start the CS-208 program and select the appropriate COM port number.
